If I add an S1 rune to my weapon, will I later be able to add an s2?
Am I required to first add an s1 in order to later add an s2?

Yes. If you want to upgrade a weapon with the second stage of a rune, it must already have the first stage, and so on. for example, if you wanna upgrade a weapon with s3, you must first have s1, then s2, then s3.
